Overview

In this role you will advance conjugation processes for antibody-siRNA conjugates, bridging discovery to GMP kilogram-scale manufacturing. You will lead scale-up efforts within the Process Chemistry team, collaborating with CRO/CDMO partners to ensure robust, quality processes. You will own optimization, analytical characterization, and tech transfer to internal and external manufacturers. This position offers hands-on bench work and cross-functional impact on Arrowhead's TRiM platform expansion into targeted extrahepatic delivery. You will help enable a pipeline of RNAi therapeutics with broader tissue targeting.

Responsibilities
  • Optimize discovery-stage conjugation protocols for antibody-siRNA conjugates to improve yield and reproducibility for scale-up
  • Develop GMP-ready, kilogram-scale conjugation processes, including site-specific and stochastic strategies, linker chemistry, and payload attachment
  • Hands-on lab execution of conjugation, purification, and analytical control strategies
  • Oversee CROs/CDMOs for process development and manufacturing, setting direction and ensuring timelines and quality
  • Characterize conjugates (DAR, aggregation, charge heterogeneity, stability) using HIC, SEC, IEX, LC-MS, AEX
  • Lead technology transfer to manufacturing partners; author process descriptions, batch records, and development reports
  • Troubleshoot process/analytical challenges; design experiments to define parameters and robustness for kilogram-scale operation
  • Collaborate cross-functionally with discovery chemistry, linker/conjugation chemistry, analytical development, and CMC teams
Key requirements
  • PhD in relevant field or equivalent hands-on experience
  • Hands-on experience in siRNA conjugation development
  • Knowledge of oligonucleotide/antibody chemistry, linker design, and bioconjugation methods
  • Familiarity with purification and analytical characterization of conjugates
