Principal Scientist, TCR Optimization

Overview

In this role you lead the development of scalable TCR optimization workflows to advance TCR-based therapeutics. You will coordinate wet-lab experiments with computational decisions, collaborating across discovery, engineering, AI/ML, and preclinical teams. You drive end-to-end optimization to improve affinity, specificity, stability, and developability for therapeutic formats. Responsibilities
  • Build, optimize, and execute scalable TCR optimization workflows using display technologies and directed evolution
  • Establish an end-to-end TCR optimization pipeline linking design, screening, characterization, and lead selection
  • Design rational and semi-rational TCR libraries informed by sequence/structure and developability needs
  • Lead multi-parameter optimization programs balancing affinity, specificity, stability, expression, developability, and potency
  • Partner with computational scientists to integrate cross-reactivity, structure, and developability data into decisions
  • Interpret complex screening and biophysical datasets to prioritize candidates and refine strategies
  • Support programs by delivering optimized binders suitable for TCR-T, TCR engager, and related modalities
  • Evaluate new display/screening/automation/protein-engineering technologies to raise throughput and quality
  • Collaborate with AI/ML, NGS, functional assays, reagent generation, preclinical, and external partners to generate actionable data
  • Communicate strategy, results, risks, and tradeoffs to multidisciplinary teams
  • Communicate scientific strategy and recommendations to project and platform teams
Key requirements
  • PhD in immunology, protein engineering, molecular biology, biochemistry, biophysics, cell biology, or related disciplines
  • At least 5 years of post-PhD experience in TCR engineering, protein engineering, directed evolution, or related fields
  • Experience executing TCR or TCR-mimetic optimization workflows
  • Hands-on experience with display technologies and library design for affinity maturation and developability optimization
  • Experience designing and interpreting binding, stability, expression, functional potency, or biophysical assays
  • Ability to lead complex experimental workflows and deliver high-quality datasets under timelines
  • Experience collaborating with computational biology, AI/ML, NGS, reagent generation, functional assays, preclinical, or external partners
  • Ability to make and communicate key decisions based on data and stakeholder feedback
  • Excellent written, presentation, and interpersonal communication skills
